Cord Blood Collection Protocol

Collect Cord Blood Sample

Instructions for Collection Prior to Expulsion of Placenta

  1. After delivery of the infant and prior to expulsion of the placenta, double-clamp and cut the umbilical cord.
  2. Sterilize the proposed needle insertion site of the cord with the antiseptic prep pad provided. The proposed needle insertion site should be just above the clamp that remains on the cord.
  3. Insert the needle of the Collection Bag provided into the cord and collect as much sample as possible via gravity-flow, aiming to fill the bag (a minimum of 80 mL is required). The cord will appear empty and pale when collection is completed.
  4. Tie two tight knots in the tubing as close to the needle as possible.
  5. Gently rotate the bag several times to thoroughly mix the cord blood with the anticoagulant.

Instructions for Collection Following Expulsion of Placenta or After Cesarean Delivery

  1. Double-clamp and cut the umbilical cord.
  2. Place the placenta and cord in a sterile tray, bowl, or Chux pad with the fetal side down.
  3. Place the placenta on an elevated surface if possible, and drape the umbilical cord out of the tray or Chux pad.
  4. Sterilize the proposed needle insertion site of the cord with the antiseptic prep pad provided. The proposed needle insertion site should be just above the clamp that remains on the cord.
  5. Insert the needle of the Collection Bag provided into the cord and collect as much sample as possible via gravity-flow, aiming to fill the bag (a minimum of 80 mL is required). The cord will appear empty and pale when collection is completed.
  6. Tie two tight knots in the tubing as close to the needle as possible.
  7. Gently rotate the bag several times to thoroughly mix the cord blood with the anticoagulant.

Label Cord Blood Sample and Complete Form

  1. Affix a Collection Kit CASE ID to the Collection Bag and place the Collection Bag into the Specimen Transport Bag.
  2. Complete the Collection Form.
  3. Start the temperature datalogger by using a pen or pointed object to depress the indented ‘start’ button on the temperature datalogger. The sample must be maintained at a constant ambient temperature to ensure maximum viability of stem cells.
  4. Replace collected sample and datalogger into the Collection Kit.

Collect Maternal Blood Sample

  1. Collect maternal peripheral blood via venipuncture into the serum (red-top) tube and the 2 EDTA (pink- or lavendar-top) Vacutainer tubes provided in the kit.
  2. Invert tubes 8 to 10 times after collection to mix.
  3. Place all three tubes into the cord blood collection kit shipping container.

After Collection of the CBU and maternal samples, return the Collection Kit to the Patient. The Patient is responsible for arranging timely delivery of the CBU to HealthCord.
